 殆どの研究者は晩年になって、自分の歩んできた研究遍路を振り返る時、そこにいくつかの重要な里標(マイルストーン)を見出すであろう。今、私か現役の研究生活に終止符を打とうとする時、私にとって重要な五つの里標のあったことが思い出される。

 まず最初は一九六二年から一九六五年における、私のミシガン州立大学農学部でのフルブライト研究員としての研究生活である。私はそれまで京都大学農学部農芸化学科大学院学生として植物栄養素の葉面吸収について研究していた。特に葉面の最外層であるクチクラ膜を酵素で分離して、そのクチクラ膜のイオン透過性を調べる新しい解析研究を行っていた。私が所属した肥科学研究室(奥田東教授)において、私は葉面吸収に関する研究をする唯一の大学院生であり、余り刺激的な研究環境でなく、もっと活発な研究討論や切磋琢磨する場所が欲しかった。そこで、私は更に活発な研究集団を海外に求める決心をした。そのため数通の手紙をイギリス、アメリカ、オーストラリアの研究活動の旺盛と思われる研究室に出した。そして最初に返事のあったのがアメリカのミシガン州立大学からで、滞在費として年額$六〇〇〇を支給するとの返事であった。私は貧しい若い研究者で渡航費がなかった。そこで、フルブライト研究員の渡航費支給の試験を受け、幸い合格し、渡航費を作った。

 これが、私のアメリカ中西部ミシガン州イーストランシング(州庁所在地)で外国の研究者と共に三年間を過ごす切っ掛けとなった。このような偶然の機会が、私の将来の研究生活に大きな視野で物事を洞察し、もっと豊かな人生観を持つことを学び、また、人にはそれぞれの独自性があり、各人異った考えのもとに生きていることに気付く機会が与えられた。この人生経験は私の科学に対する心のみならず、人間としての生き方に大きな影響を与えたことは間違いない。

 当時、私の研究の興味は葉面から分離したクチクラ膜と酵素的に葉肉組織から分離した柔細胞を用いて、人工的な葉のモデルを構築することであった。そのため種々の酵素を用いて葉肉組織から柔細胞を単離し、クチクラ膜とあわせて葉のモデルを再構築し、そのモデルを用いて養分吸収を研究することを開始した。しかし、単離した柔細胞への徹生物の混入をどうしても避けることが出来ず、このシステムによる葉面吸収の研究は失敗した。

 この時点に於いて、私の研究の発想は大きく転換した。即ち、多量の無菌葉肉細胞を得るために、無菌下で葉肉組織の培養の研究を開始した。これが後に、佐藤文彦(現京都大学教授)らの光独立栄養細胞培養の成功へと展開していく。即ち、通常の植物細胞培養では炭素源を糖類に依存する従属培養であるが、一方、光独立栄養細胞では、自己の葉緑体で光合成を行い、葉肉細胞と同じような機能を発現して増殖している。
 一九六五年に京都大学農学部に復職したが、既に私の研究上の興味は植物細胞培養に完全に移行していた。帰国した頃は植物細胞培養を行っている研究者は我が国では極めて少数であり、それぞれが個々に自分の興味ある植物の細胞培養を試行していた。当時、世界で単子葉植物イネ・コムギなどの主要穀物の組織培養は極めて困難とされ、未だ成功していなかった。そこで私達のグループは直ちにイネやカラスムギなどの組織培養とこれらの培養細胞からの個体再生への研究に取り組み、成功した結果をネイチャー誌に二報報告した記憶がある(西豊行、現日本新薬一場志埼、現大塚製薬研究課長)。この様に新たな研究領域への転換を私の第二の里標として見ることが出来る。

 振り返ってみると、もし私がアメリカで勉強していなかったら、アメリカ的なチャレンジ精神を身につけることもなく、おそらく生涯、葉面吸収の研究に精力を尽くしていたであろう。一九六〇年初めは、ケネディー大統領の大々的な科学振興政策で、アメリカの科学研究が大変活性化した時代にアメリカに滞在できたことも幸いであった。

 日本の国立大学は当時講座制として、一教授、一助教授、二助手から構成されており、数授か退官しない限り、助教授は教授に昇任することが出来なかった。一九六七年に私は助教授となったが、教授とは四歳ちがいで、私がこの講座で教授になることは殆ど不可能であった。しかし、一九八二年文部省と京都大学農学部教官の御尽力で生物細胞生産制御実験センターと称する植物細胞培養研究を中心とする農学部附属施設が一○年時限で設置された。そして、私はそこの教授となり全力を挙げて植物細胞培養の研究に熱中した。この一〇年時限の研究環境は当時五〇歳の自分が全力を尽くす背水の陣を敷く機会となり、大変よかったと思っている。

 一九七〇年中頃から、私達の研究グループ、佐藤文彦、渡辺克美(現近畿大学助教授)、山本好和(現秋田県立大学教授)らと共に、私は植物培養細胞が葉緑体形成や二次代謝物質産生などの機能発現に関して、極めてヘテロであり、その結果培養細胞は機能発現に関してモザイク的構成をしていることを発見した。その結果、目的とする機能発現の研究を行う場合、より機能を発現している細胞を選抜することの重要性に気付いた。特に、有用二次代謝産物の大量生産の基礎から応用への研究に興味を持った私はアントシアニン、イソキノリンアルカロイド、シコニン、トロパンアルカロイドなどを高産生する細胞の選抜に集中した。これは将に植物バイオテクノロジーの幕開けであった。この細胞選抜という操作の繰り返しによって、高産生細胞株を確立し、ジャーファーメンターによって有用物質の大量生産系を確立することが出来た。この細胞のモザイク構成の発見まで、植物細胞培養に関わる研究者は殆ど培養細胞は均一な細胞集団と考えていたというのが事実であったと思う。

 この植物培養細胞の機能発現に差異があることの認識に基づいて、私達の研究はアントシアニン高産生株、シコニン高産生株、ベルベリン高産生株、アトロピン高産生株などの細胞株を選抜培養することにより、植物細胞培養による有用物質生産の植物バイオテクノロジーを世界に先駆けて展開することが出来た。この研究展開の基礎研究には文部省の科学研究費、特に特定研究の継続的支援が大きな力となったことを記しておきたい。さらに、化学工業関連企業が私達と協同研究を実施し、企業レベルにおける工業生産にまで発展させた。一方、私たちは特定有用二次代謝産物の高産生株を得ることが出来たので、それぞれの二次代謝産物を生合成する代謝経路ならびにその生合成に関与する酵素反応機作の研究に着手した。さらに、生合成の酵素化学反応の基本となる酵素の遺伝子の単離、ならびにその遺伝子の発現機構など分子生物学の手段も取り入れた。特に中心となった代謝経路はイソキノリンアルカロイドは佐藤文彦、杉本幸裕(現神戸大学教授)、トロパンアルカロイドは橋本隆(現奈良先端科学技術大学院大学教授)を中心としてBirgit Drager(現独、ハーレ大学教授)、Edward Leete(故人。ミネソタ大学教授)、Dae‐Jin Yun(現韓国Gyeongsang国立大学助教授)、中島敬二(現奈良先端科学抜術大学院大学助教授)、遠藤剛(現京都大学講師)、星野祥子(主婦)らの協力で殆どの代謝経路を解明することが出来た。これが私の研究遍路に於ける第三里標であり、如何に困難な環境であっても、努力するといつか道が拓り開かれることを教えられた。

 一九九一年第四の大きな里標を迎えた。当時関西文化学術研究都市に我が国にとって新しい型の大学院のみの大学を設立する計画があり、私もその設立にあたって委員となっていた。私達はこの新しい大学院大学の教員、学生などの構成についても柔軟性をもったものとし、教育・研究のいずれにおいても国際的に第一流の学府にしたいと考えた。勿論、教員は国家公務員であるが、その給与体系は年功序列に従わないで、実績評価において決定し、人材は国内外に求め、且つ一講座の構成は教授一、助教授一、助手二とするが、これは教育、研究の単位とするのみで、講座間の壁を取り除き、講座のセミナーも他講座の学生達も参加できるなど、旧来の大学院制度の枠を打ち破るシステムを構築することを目指した。実際には当初の理想は多くの法的規制などにより仲々達成させることは出来なかったが、少なくとも従来の旧帝大系大学の様に長い歴史と伝統に基づいた閉鎖的な大学管理運営の学府でない大学院大学が創設された。一九九一年一〇月に情報科学、バイオサイエンスの先端科学分野に焦点を当てた大学院大学の設置が決定され、一九九三年情報科学研究科、一九九四年バイオサイエンス研究科の学生を受け入れた。

 私にとって新しい形式の大学院大学の創設は非常に興味はあったが、このような大学こそ若い教員の力で築き上げるのが一番よいと考えていた。しかし、奈良先端科学技術大学院大学と京都大学の強い要請もあり、一九九四年に私は新大学へ移籍することにした。京都大学でともに研究してきた仲間を二分して、研究室の仲間に助けられてバイオサイエンス研究科に新しい講座を開設した。新しい大学では残された年月を研究に集中するため、一切の管理職を辞退したが、最後に遺伝子教育研究センター長だけは併任した。研究としては従来からのトロパンアルカロイドの代謝反応の集大成として主要代謝反応酵素の結晶化と酵素の高次構造の決定などの研究を継続した。

 一九九七年に私は第一回の学長選挙によって学長に選出された。本学の学長任期は四年一期のみとされていた。私は新大学に移る前から前立腺ガンにかかっており、四年の任期を全うできるか不安であったが、学長就任を受けた。これが第五の里標である。同時にこの様な発足直後の新大学では大学管理運営と自己の研究を両立させることは極めて困難と認識した。その結果、少なくとも学長在任中は大学の管理運営に集中する決意をして行動した。在任中、入院加療して、大学に御迷惑をかけたこともあったが、大学教職員全員と京大医学部の先生方の御支援で四年間の任期を大学の発展に寄与することが出来た。第五の里標を終えて、私は三六年にわたる教育研究に充分打ち込ませて頂いたのみならず、私にとって非常に不向きと考えている大学管理運営にも四年間たずさわる機会を得たことに感謝している。そして、管理運営の業務に携わる者にとっても自分の過去の充分な教育と研究経験が非常に重要であると思う様になった。
